Das Museum für byzantinische Kultur zielt darauf ab, verschiedene Aspekte des Lebens in der byzantinischen und postbyzantinischen Zeit zu präsentieren: Kunst, Ideologie, soziale Struktur und Religion sowie wie sich historische Veränderungen und die politische Situation auf den Alltag der Menschen auswirkten.
Gleichzeitig wurden die Tätigkeit der Abteilung für Bildungsprogramme, die gute Struktur und Funktion der Konservierungslabors und der Lagerräume für archäologisches Material, die Bereitstellung von wissenschaftlichem Know-how für andere Balkanländer, die Organisation wissenschaftlicher Treffen und Konferenzen sowie sowie die Redaktion und Veröffentlichung machen das Museum zu einem außerordentlich wichtigen Zentrum für die Erhaltung, Erforschung und Förderung der byzantinischen und postbyzantinischen Kultur. Seit der Einweihung des Museums im Jahr 1994 erscheint ein jährliches Bulletin, das erste seiner Art von einem griechischen öffentlichen Museum.

Das Museum befindet sich in einem vom Architekten Patroklos Karantinos entworfenen Gebäude und ist ein Beispiel für die modernen architektonischen Trends Griechenlands. Das 1962 erbaute Museum wurde 1980 um einen neuen Flügel erweitert, in dem die Funde aus Vergina bis 1997 ausgestellt waren. In den Jahren 2001 und 2004, im Vorfeld der Olympischen Spiele 2004 in Athen, wurde das Museum umfassend renoviert und seine Dauerausstellungen neu organisiert.

In den zentralen Räumen befinden sich Exponate aus den archäologischen Ausgrabungen in Thessaloniki und im weiteren Bereich Mazedoniens. Der neue Flügel beherbergt zwei Ausstellungen: Das Gold von Macedon, mit Artefakten von den Friedhöfen von Sindos, Agia Paraskevi, Nea Filadelfia, Makrygialos, Derveni, Lete, Serres und Evropos; und das Gebiet von Thessaloniki in der Vorgeschichte, mit Material aus prähistorischen Siedlungen, Dating vom Neolithikum bis zum frühen und Späte Bronzezeit.
